Be cautious if your wash buffers contain Tween 20. Our IHC protocol uses wash buffers containing 0.3% Tween-20 (TBST). Despite the claimed chemical resistance of up to 1% Tween-20, the hydrophobic barrier no longer works after 15 minutes of washing. This is still the best deal for a PAP pen out there, but before performing your actual staining, definitely test the pen against your wash buffer on a blank slide.
