Be cautious if your wash buffers contain Tween 20. Our IHC protocol uses wash buffers containing 0.3% Tween-20 (TBST). Despite the claimed chemical resistance of up to 1% Tween-20, the hydrophobic barrier no longer works after 15 minutes of washing. This is still the best deal for a PAP pen out there, but before performing your actual staining, definitely test the pen against your wash buffer on a blank slide.

Store owner replied: Thank you Jack for your review and comments, we've been trying to replicate the issue. I believe that the problem comes from the application of the PAP ink on semi-wet slides, as is the case of application after the wash step after antigen retrieval. Our workflow has been to dry the slides completely before drawing with the PAP pen. When we've left the slide semi-wet, the ink is still applied but gets washed away. Please see if this helps your workflow!
